A recurring trend in Batman shows is to primarily use Batman's main enemies, while throwing in a few new villains. The 60s' Batman show created several of their own villains.
Villains That Were Originally From The Comics:
The Joker (In most versions of Batman, the Joker is Batman's main enemy. However, in the 60's show, he and Penguin had the same amount of appearances. Also, Penguin was the one who called the shots in the 60s' movie.)

The Riddler (Riddler was actually a minor Batman villain in the comics. Thanks to how great the Riddler was in the 60s' show, he's become one of Batman's most prominent villains.)

The Clock King (Clock King has served as both a Batman and a Green Arrow villain. Clock King's appearance in the 60s' show was written by Batman's co-creator and the original writer of the comics, Bill Finger.)
False Face (False Face was a very small part of the Batman rouges gallery in the comics, but he did manage to appear in the 60s' show)
The Puzzler is the name of a Superman villain. The Puzzler was used on the 60s' show, during the time where Frank Gorshin didn't want to appear on the show, so the Puzzler was used instead of the Riddler.
Two-Face was going to appear in the show, but his appearance got cancelled. Thankfully, Two-Face got to be the main villain of the Adam West animated film, Batman vs. Two-Face.
Louie the Lilac has a flower-themed gimmick, which reminds me of Poison Ivy. Poison Ivy first appeared in the comics, during the time the 60s' show was being made, so I'm not sure if Louie the Lilac was based on her. Although Scarecrow never appeared, there's an episode where Shame the cowboy uses fear gas on Batman.
All of the other villains, that appeared in the 60s' show, were created for the show, not the comics. Although King Tut was one of the show's most recurring villains, he was a creation of the show. Same thing with Egghead. Despite that, they have made cameos in several forms of Batman media.
