1.- Physical attraction.
Cuddy likes Lucas. He is a good looking guy and she is into him. It’s that simple

2.- Lucas adores Cuddy
You can see how Lucas is always checking Cuddy out. He is not afraid to tell her how beautiful is to him. Also keep in mind that we didn't see how did Lucas wooed Cuddy.

Lucas [to House]: I don’t want just do her
Lucas [to Cuddy]: I’m into you because you’re hot, and smart…
3.- Lucas understands Cuddy (and he has the patience of saint)
Let’s face it. Cuddy can be a walking contradiction sometimes, she has issues with her job, her family (maybe), House, Wilson…Any other man would give up (like Don in 3x14 “Insensitive”), but Lucas understands Cuddy and her crazy ways.

Lucas: Look our relationship is gonna be about House or it's gonna be about us. I vote for it being about us.
Cuddy : Well, House is gonna make it about him for us.
Lucas: This is very insulting. You're apparentntly shocked by this which means that you thought there was some way that we could date, get married, have two kids, a dog, retire to Florida all without House finding out. Or you thought that we wouldn't date all that long. The first means you're delusional. The second means I'm delusional. Technically, it could also mean you thought House had matured, but then we're back to your being— (makes cukoo whistle and signs at his head)
Cuddy: Shut up. You're right. I'm sorry.
Lucas: I'm also flattered. Yeah. 'cause you acted all cool and everything with Wilson, but you feel comfortable enough with me that you can freak out.
4.- Lucas is good with Rachel (And Cuddy trusts him)
Rachel is the most important person in the world for Cuddy, and she trusts Lucas to babysit for (what it seems to be) long periods of time. Lucas must be good with the little girl, he earn that trust.

Cuddy comes back form work to find Lucas and Rachel playing. I’m gonna assumed that Lucas babysit all afternoon (And he had time to cook dinner)
5.- Lucas is weird.
Cuddy wants a taste of a normal life, but, can you imagine Cuddy with a jewish lawyer? A widower accountant? A Divorce architect? Too boring for Cuddy. But Lucas is just weird enough. He is the safe choice, without being the boring choice.
He has an odd job, he has a peculiar way of dressing, and a complicated thought process, and yes he can be a little creepy sometimes.

Lucas: I think its a good idea to get to know a little bit more about a woman before you ask her out. Show that you care about more than just what she looks like.
Cuddy: As romantic as you make that sound, I'm pretty sure that what you're doing is not so much caring as creepy.
Lucas: That's not fair.
Cuddy: You were scrounging through my desk.
Lucas: What the hell am I going to find out? You went to Michigan. You like your sandwiches a little light on mayo. You stay in touch with your mother. You rented The English Patient. And you gave money to Amnesty International. There's not one thing I can find out in here you wouldn't tell a coworker in an elevator. I'm not a creep, I just thought you seemed interesting.

Okay I have no evidence of this, but you know it’s true.
For all those reasons I do believe Cuddy has developed feelings for Lucas
